概要
Aging is characterized by cognitive decline and metabolic abnormalities. Lipids with palmitic acid at the β-position (sn-2 palmitic acid), which are abundant in breast milk fat, regulate inflammation and lipid metabolism, but their effect on aging was unclear. In this study, aging-accelerated mice (SAMP8) were administered Sn-2 palmitate-enriched lipids for 8 weeks. As a result, cognitive function improved, and hippocampal aging markers (p16, p21) and inflammatory cytokines (IL-6, TNF-α) decreased. Furthermore, the NAD⁺/NADH ratio and Sirt1/3 activity increased, and mitochondrial biosynthesis-related factors (PGC-1α, NRF1, TFAM) increased. This suggests that Sn-2 palmitate-enriched lipids may mitigate aging-related cognitive and physiological declines through NAD⁺ metabolism and mitochondrial function.
